Alterations in the permeability of Neurospora crassa due to polyene antibiotics.

چکیده
Kinsky, Stephen C. (Washington University, St. Louis, Mo.). Alterations in the permeability of Neurospora crassa due to polyene antibiotics. J. Bacteriol. 82:889-897. 1961.-Thirty-three antibiotics and synthetic fungicides were examined for their effect on the growth and morphology of Neurospora crassa. Only the polyene antibiotics (nystatin, amphotericin B, and filipin) caused a decrease in the dry weight of mycelial mats, which was accompanied by the appearance of cytoplasmic constituents in the medium. The influence of polyene concentration, incubation time, mycelial age, various metabolic inhibitors, pH, and media composition on this phenomenon are described. The present evidence indicates that the polyenes exert their primarily fungicidal effect by an alteration of permeability, probably due to direct action on the membrane of sensitive organisms.

متن کاملPenetration of Lipid Monolayers by Polyene Antibiotics. Correlation with Selective Toxicity and Mode of Action.
Previous studies have shown that polyene antibiotics alter permeability in sensitive fungi and thus lead to the loss of essential cytoplasmic constituents, which ultimately culminates in cell death (l-3). Subsequent investigations have led to the contention that the selective toxicity of the polyene antibiotics is due to interaction with a component present only in the membrane of sensitive org...

متن کاملMembrane permeability and the loss of germination factor from Neurospora crassa at low water activities.
Neurospora crassa conidia incubating in buffer at low water activities (a(w)) release a germination-essential component as well as 260-nm absorbing and ninhydrin-positive materials, regardless of whether an electrolyte or non-electrolyte is used to reduce a(w). Chloroform and antibiotics known to increase cell-membrane permeability have a similar effect. This suggests that membrane damage occur...
متن کاملBiotransformation of Hydrocortisone by Neurospora crassa
The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...
